Résumé : Preterm delivery (J. Biol. Chem. 2010, 285, 25624-25636) and aza-amino acyl proline analogues (Bourguet, et al. J. Med. Chem. 2011, 54, 6085–6097) can reduce prostaglandin-F2α-induced myometrial contractions and modulate prostaglandin F2α-induced G-protein signaling pathways. My presentation will discuss advances in understanding the relationship between structure and activity of aza-amino acyl proline modulators. The biological activity of these modulators is contingent on their aaa-residue side chain. Diversity-oriented synthesis methods to introduce different side chains onto the aza-amino acyl proline modulators have been pursued featuring alkylation of aza-glycinyl- proline analogs, as well as copper-catalyzed reactions on aza-propargylglycinyl residues. Finally, photo-reactive modulators have been pursued to tag the FP receptor and identify the allosteric binding site.
